Ultra Sound Assisted Extraction of Aminoacid, Heterocyclic Compounds and Guanosine Rich Fraction of Glycyrrhiza glabra and its Invitro Antidiabetic and Antilipidemic Activity

Abstract

The present study was aimed to assess the antidiabetic and antilipidemic activity of G.glabra extract. The root powder of G.glabra was extracted with a mixture of ethanol, methanol and acetone at 1:1:1 ratio under ultrasonic waves at 4°C for 15  min. The extract was characterized by UV-Vis spectroscopy, FTIR and GC-MS. The extract was assessed for its antidiabetic activity through the inhibition of porcine pancreatic amylase activity. IC50 of the extract to amylase activity was assessed from 100 to 500 µg/mL. The mode of inhibition of amylase by G.glabra extract was evaluated by enzyme kinetics assay. The reducing sugars released by amylase activity was quantified using dinitrosalicylic acid. Inhibition of amylase activity was calculated from the control tubes incubated without enzyme in the reaction mixture. UV-Visible spectrum, FTIR and GC-MS confirms the presence of amino acids, heterocyclic compounds, guanosine etc. IC50 of G.glabra extract to porcine pancreatic amylase was 185µg/mL. The extract showed mixed type of inhibition with decrease in both Vmax and Km. The inhibition lines intersect in the X axis in the second quadrant confirming the mixed type of inhibition.
